چکیده
Specific PCR products are detected with an antibody-free lateral-flow device by sandwiching them between reporter oligonucleotides covalently attached to gold nanoparticles (GNPs) and capture oligonucleotides covalently attached to a nitrocellulose chromatographic strip.

Rapid Molecular Detection of Multidrug-Resistant Tuberculosis by PCR-Nucleic Acid Lateral Flow Immunoassay
Several existing molecular tests for multidrug-resistant tuberculosis (MDR-TB) are limited by complexity and cost, hindering their widespread application. The objective of this proof of concept study was to develop a simple Nucleic Acid Lateral Flow (NALF) immunoassay as a potential diagnostic alternative, to complement conventional PCR, for the rapid molecular detection of MDR-TB. The NALF dev...
متن کاملOligonucleotide-linked gold nanoparticle aggregates for enhanced sensitivity in lateral flow assays.
Lateral flow assays (LFAs) as rapid analytical techniques promise to be widely used in point-of-care (POC) diagnostics because of their affordability and simplicity. However, LFAs still suffer from low sensitivity in detection of various biomarkers, e.g., nucleic acids. In this study, we developed a simple and general one-step signal amplification strategy, which employed oligonucleotide-linked...
متن کاملSimple System for Isothermal DNA Amplification Coupled to Lateral Flow Detection
Infectious disease diagnosis in point-of-care settings can be greatly improved through integrated, automated nucleic acid testing devices. We have developed an early prototype for a low-cost system which executes isothermal DNA amplification coupled to nucleic acid lateral flow (NALF) detection in a mesofluidic cartridge attached to a portable instrument. Fluid handling inside the cartridge is ...
متن کاملUV Treatment Nucleic Acid Probe Without Biotin-Labeling Is Sensitive and Sufficient For The Fabrication of Nucleic Acid Lateral Flow (NALF) Strip Test
Nucleic acid lateral flow (NALF) test strip is currently a promising method in biomedical applications for point-of-care diagnostic tests. Generally, biotin-labelled probe is commonly used in making the test lines and control lines, and the entire processes require long periods of incubation time with the probe and streptavidin. In this study, we use non-labelled oligonucleotides that have been...
متن کاملElectricity-Free Amplification and Detection for Molecular Point-of-Care Diagnosis of HIV-1
In resource-limited settings, the lack of decentralized molecular diagnostic testing and sparse access to centralized medical facilities can present a critical barrier to timely diagnosis, treatment, and subsequent control and elimination of infectious diseases. Isothermal nucleic acid amplification methods, including reverse transcription loop-mediated isothermal amplification (RT-LAMP), are w...
